I was thinking about making a mod overhauling sexuality and attraction opinion, in which the homosexual trait is replaced by two new traits: "attracted to opposite gender" and "attracted to same gender". The attraction opinion would be modified to only affect spouses, lovers, and seduction targets who have the appropriate trait. Is this modification of attraction opinion possible?

Hmm, a good idea.
However, the Homosexual trait is pretty hardcoded, and afaik you can't use any other trait to enable/disable the effects from "sex_appeal_opinion".
You could probably set up a new layer of traits over the current framework and have them interact, but the effects from normal traits would be unaffected.
So basically what you want to do is impossible because hardcoded :(

Is there a general way to specify "affects opinion, but only if X trait is present"? I know there are ways to specify it for someone who has the same trait, or the opposite trait, or is the same religion, or the same dynasty, or is a spouse.

Not that I know of.
Only those you name, so you could set up some opposite traits that generate an opinion towards each other.
But nothing complex like you want, with more than two things interacting at any time. The most complex thing vanilla allows is probably same_opinion_if_same_religion, but that is hardcoded and you can't add others in that style.

It's a shame, really.
