As a newb I find myself building bus routes by default. I can't find much information on what the best use of each vehicle type is, other than metro which seems to be more for long distance routes.

What characteristics of a route make it more suitable for trolleys, trams or buses? The tram/trolley thing is the one I can't really get my head around. Are trams for long routes or short? Far apart stops or close?

Is it just about how fast you want them to get around and how many people they carry? I.e. a tram is good to get more people a bit further a bit quicker?

I suspected, but wasn't sure, that perhaps you're supposed to upgrade as things get busier, e.g. start with a bus route and upgrade to trams if its successful maybe.

Any hints more than gratefully welcomed!

Trams can go in the middle of avenues where there are no cars, so that's a plus.
And I don't know, but I generally do find my tram and trolley routes more profitable than bus ones. I assume the original track building investment pays off overtime, as it's somehow more popular?

Like the first City in Motion, metro means instant win. No traffic problems, and earns you the most. It is clear, however, that they have tried to downplay their importance. (Or like others have said, they simply did not release a complete product.)
