Do I need to buy all 34 during one game or just buy 34 among all the scenarios that I have played in the game to unlock the vehicle collection achievement? If it's the first case, it will probably take me forever, since one game year last about 1 hour in the fastest setting.

I suppose you should own all of them during a single game. To make the waiting shorter, you can already start in 1945 (all the vehicles that appear in 1920 are still available in that year, including the first bus Maurice Astor).
After letting it run for a day, I'm in 1955, not building anything since it will slow down the game, 50 more years to go.. I do have a huge budget surplus though. .hah
it's about 2.5 minutes a month, so they best my computer can do is 2 year / hour, so that's 25 hours to go.
