Yes, literacy DOES affect pop promotion.
15% is the magic treshold for literacy - if you are below that, you receive a -0.5 penalty to pop promotion chance.
Every 5% above 15% (up to 80%) will grant additional promotion benefits. The faster you hit 15%, the fater you can improve your country. And remember that the benefits from early improvement becomes exponentially large in the late game.
I had a RUS game yesterday and rushed biologism asap - turned out to be a good move.
Another benefit is that literacy increases CON - and as long as you don't want to stay an absolute monarchy, that is a good thing.
My personal tech priority for RUS was:
(1) RP increasing techs.
(2) Industrial and commerce techs until my RGOs make decent profit.
(3) Plurality increasing techs (even more RP!).
(4) National Focus techs.
(5) RGO improving techs.
(6) Commerce organization techs to improve factory input and output (Management Strategy!)
(7) Industrial Throughput techs.
