Ah, yeah, forgot we have the FAQ.

So.... if a total of 9 tiles gives about 36 km2 to work on..... and if we unleash the full potential of the tiles to (about?) 16, we'll get... 64 km2 .... and the city I'm working on would probably need about 400 km2, making the scale .... 1 to 6.25 (which is quite reasonable), and making the in-game population count to work towards 480,000 people.... Seems reasonable.....

With modding you'll be able to build on all 25 buildable tiles, giving you a 100km² square (no guarantees your pc won't explode though, and no technical support when you do this mod). There's also 2 tiles in every direction for horizons/eyecandy. So you'll be able to build on 10x10km from a 18x18km map.

According to the CEO in the AMA, that's no longer the case which saddens me to no end. It had been said previously that you could.

EDIT: It seems there is confusion as to whether you actually can or can't.
EDIT 2: Yeah, it was a misunderstanding of the question from what I'm able to tell and yes, all 25 tiles will be able to be unlocked with modding.
