No you can't. The direction of the one way road is determined by the start and end point when you build it.

Yes, it's cumbersome because if you draw a box and then want to expand it to a rectangle with the one ways going in either direction as the longer lines, it's almost impossible to line up the inbound road properly.
I've literally only spent a few game minutes in this so far, but I'm already pretty happy with the game overall. I've been a huge SimCity fan since the first one and was pretty disappointed with their latest iteration. I'm glad Paradox is moving into this niche because with EA shutting down Maxis, the genre is going to need their unique fan commitment.
The mod that cordo links to is just the thing you want - You will be able to drag the direction of roads to upgrade them to one ways.
