I played a ton of Ck2 and am just starting Ck3 - One of the things you needed to do in ck2 was to get your courtiers married, and then to educate all the children in your kingdom. Do you still have to do those two things in ck3?
Not sure what you mean by "need" to, but yes it is still possible to do all of these things. OK, I'll set guardians for all the children and check every few years and get wives for my knights...
Now how about spending money? Is it better to build new holdings or buildings? Generally in ck2 I prefered to build cities, at least on my ocean front counties, for the cities bonuses once my economy got going. Is it the same here or are buildings a better option? Playing as the King of Leon I see upgrade hill farms for $200 and get $.5 tax/month or build a new holding for $400 which seems a better deal.

This is correct. A target which is not a ruler themselves awards Prestige like their liege, if they are married to or Close Family of that liege.Do you get more prestige from romancing someone of a higher rank, e.g. the wife of a king vs. the wife of a count? From what I've seen this seems to be the case, but I'm not sure.
if your eldest son is dead, his eldest son takes his place in the inheritance chain. So, your 1st son's 1st son's 1st son will be your player heir, and then your 2nd son (or his descendants) gets the next share of the partition, etc.How does title inheritance skipping generations work?
