They won't drain a river because the source is constantly causing it to refill, but if you manage to section off part of the river into a makeshift-lake, then yes, pumps will cause it to go dry eventually.

Actually they can and they WILL if the river isn't spawning enough water - A lot of smaller rivers can dry up with just a few water pumps easily.
Pumps suck in water - Sewage Outlets pump it out. You can even pump out your sewage into a Hydro Dam resevoir to help it generate more energy (provided the sewage won't eff up your pumps - But you can always switch to water towers)
