What affects diploannexation?
Definetely I think:
- # of provinces I own,
- # of provinces of the target counrty,
- diplo score,
- stability (? - not sure about it),
- relation score,
- years of vassalisation.

I wonder if there is some other factors I ignore to enhance the DA probability, as I really want to DA a vassal I have had for more than 50 years, but maybe I'm too small to do that (me 6 provinces, they 3), even though I have a 6 diplo rating, +200 relation and +3 stability.

Income is also a huge factor in DA'ing other countries.
And the size of your army matters as well, you need a lot bigger army than they have. The sword is, in this period, migthier than the pen.

Diplo Annexing usually involves a lot of luck as well, sometimes they accept the first time sometimes they accept the 10th time, it usually depends on your patience.

Otherwise just get into a war that will destroy your allies army and pick up a few provinces yourself, should gbe enough to make them accept. In my experience the AI usually accept to be annexed after a war rather than before.
