NOTE: I want it to last; i.e. so that immediately afterward Persia or the Ottomans will send another colonist and change the culture back.

Should be more than easy.

But I want it to specifically trigger when it is at a certain population. Otherwise it would be redundant.

In the start of the savegame there is a list of all provinces, like this:
province = { id = 67 whiteman = yes }

You can change province culture here by adding:

province = { id = 67 whiteman = yes culture = "culture" }
