well ,what are all the method's that i can apply to maximize migration to specific colony's with future statehood in mind? (apart from letting population grow by medicine and healthcare for ex)

There isn't really much you can do. You can expand the RGO and hope it attracts immigrants, but that's about it. If you really want to grant it statehood, you could fond a province that's close to having a majority of state culture pops, convert the non-state pops to soldiers, and march them off to die. Not a nice thing to do (and not something I use), but it should be possible.

Is an RGO's attraction greater if it has more unused slots? If, say, I have an RGO level 1 with 4 POPs employed and one slot open, vs. another RGO level 3 with 8 POPs employed but with 8 slots open, is that second RGO more likely to see immigration?

First thing you'd want to do if not already done is change Malthusian thought. Currently it gives -50 to migration to colonies. Just remove the - and that'll help. Otherwise, just leave your RGO's at home unexpanded and when pops split >gonna be a long long time depending on who you are< you should hopefully have some of them traveling over seas.
