In the iron century start there is a Han realm west of the Protectorate called Guiyu. The Protectorate is represented as Uyghur but Guiyu is Han and independent. Is there any actual historical evidence for this Han enclave near the Taklimakan Basin in those years? If so is the ruling Cao dynasty of relation to the rulers of the state of Wei in the third century?

It's actually called out in the description of the iron century bookmark:

Holding control over the Guiyi Army - the last Tang presence in the West - Cao Yuande plots to build a new kingdom for himself away from the influence of his wartorn homeland.

A quick search brings up this Wikipedia article. No doubt you can continue looking from there...

EDIT: Re relationship with the famous Cao Cao, my tentative answer based on a quick skim and general reasoning is "probably not". (It's a one-syllable family name, therefore presumably fairly common, and it's far away both in space and time. And with the recent hype around 3 kingdoms, I'm sure some wikipedian would have added a relationship if there was one there.) Of course, they might be related in the biological sense even if there's no direct patrilineal descent - 7 centuries is a lot of generations.

Interesting, I think I know what my next campaign is...
