Of course, this trailer doesn't show real gameplay, but you can see that the water freezes in winter. Which is pretty cool. I hope that it will somehow affect river vessels if it is actually implemented

He said 2021 ( here ) so I'd expect development to have started in 2019, maybe 2020 at the latest.
That's what I'd say based on their earlier interviews, and some scant information that kept appearing here and there, including a meeting with modders in summer 2019 most likely (must had been before pandemic), which sounded very interesting back then, and some information released around announcement for the Airports DLC (so end of 2021) that the map would be enormous.
All we know so far is that the development could have started way before 2019 - so way before anyone could try using Unreal Engine 5.
Now that we know Unity 5 has made huge progress since it was released... - We'll see what happens ;)

And yes, although the trailer is said not to be the actual game footage - as someone pointed out (and probably few other people) it would amount to deceiption if a trailer said one thing about the game and the game was completely different.
What I'd focus on is the things shown there and what mechanics they may be indicative of ;)
yes I liked the visuals... and aparently, they could be very similar in a game using Unity 5 too. The only worry is, regardless of what any youtubers say about Unity 5, that it still might be as flippant as the previous was.
But as we can tell now - CO had money and took their time this time (very wisely) to develop the new game together with tweaks to the game engine I suppose. This is all I wanted all this time :)

Going back to the features - mixed use buildings...
What we've seen in the trailer is so far exactly the same as in the Cities Skylines 1 - nothing indicating that CO has overcome the problem of buildings having a mixed use in real life - mots of commercail buildings in CS1 are simply residential buildings with slightly different ground floor and some props here and there to look like a shop.
The same happens in the CS 2 trailer... but...

But... as i said, showing something in the trailer, regardless if it's gonna look the same or different in the game (visually), is more of indicating what new might be in the new game. And they wouldn't be showing all those little details for no reason. Only question is how many new mechanics will we get :) and if they are showing buildings with shops on the ground floor... it's quite likely for a reason, but not a definite feature of the game.

But only thing to solve is just make list of agents attached to building, instead of only one. And rework building's interface. Logically it's simple, So, in addition to new atmosphere of uhm... seriousness.. this mechanic seems correct and worth to add.
