I see you can't set buses to stop at only a few stops on a given line. So to hit the stops that are really busy, I built a new line. It goes where the first one does, so the question is this: Will the same passengers spread out over the two lines since they go to and from the same place? (I know that the passengers going to/from the other non-express stops won't.)

I think no. Every line have new passengers. So if you have crowded station if you build new line which stops in the same station you will only get more passengers in the station. But i am only talking about my experience.

I've made express lines, and they become RIDICULOUSLY overcrowded with the metro. Like before I had them, there was already too much crowding, but then once I made the new express lines, a lot more people started using them. xD

express buses are fine you must build only 2 at a time becusae as the first express bus line becoame crowded then start the second one I have only 6 express lines
in one of mine cities and they work with out over crowding , I and I use only 3 metro lines that covers most of my city.

I tried to have two buses lines that connected through two consecutive stops to allow for efficient passenger exchange. And I noticed something very interesting: some people would get out from bus line 1 at the first stop, take line 2 to the next stop, and hop back in line 1. Apparently something makes passengers prefer change lines instead of just staying on the bus that goes where they want to go.

CiMfolk decide to make a journey, then they pick a method of transport to get there. They can walk, take a taxi, drive their own car, or use your Lines. Once they plan their journey, they don't ever change their plans.

If you build a Line (let's call it Line 1), which goes from A to B to C to D and back again, people who are travelling between A,B,C and D can decide to use Line 1 for their journey. Suppose Line 1 gets very busy and there's 150 people waiting at A, and 100 of them are going to D.

If you build an express Line 2 which goes between A and D only, none of the passenger waiting for Line 1 will notice. They decided, before they began their journeys, to use Line 1 and nothing can make them change their minds.

But the NEW passengers, who start their journeys after you've built Line 2, can decide to use Line 2 to get from A to D. I'm not sure if they will guess which line is quicker, or if half will choose Line 1 and half will choose Line 2.

If you really want to get rid of that big crowd of people stubbornly waiting for Line 1 to take them from A to D, there is a solution. Remove stop A from Line 1, then put it back in again and restart Line 1. Everyone waiting at A will magically teleport away, and then begin new journeys where they can choose to use Line 2 if it suits them.
