I have a question regarding diplomatic annexation as a whole.
I am currently playing Irish minor nation, and I am in process of colonizing America, but... I got into Personal Union as a junior partner of England. So, my question is concerning the diplomatic costs of annexation, more precisely, are my colonial states factored into the annexation cost, or do I need to get out of the union ASAP?

If by "colonial states" you mean Colonial Nations (that formed after you cored 5 provinces in a CN region) then, like PhoenixG said, no, they are not counted.

If the provinces haven't been included in a CN, however, they should be counted in diplo annexation cost I think.

Also, if you're playing on the latest patch (1.24), diplo annexation costs are based on whether provinces are full-stated (8 DIP/dev) or not (4 DIP/dev). This wasn't the case in 1.22 where everything was 8 DIP/dev. If you want to slow him down, full-state everything you can and develop.
