Do you already have a city next to it, or is it on the coast? Those used to be prerequisites for you to be able to create colonies.

He already has a colony.

Most likely scenario is that you do not have an available colonist or not enough money.

The higher Trade level you have the higher is the success of placing a Trade Post (TP), a higher your monarch's administrative skill the higher is the success rate for placing Colonies.

Also conquistadors will increase the chances, for colonies with ports a stationed Explorer in the port will also increase your chances.

My guess would be it's a province that doesn't meet requirements for sending a colonist (ie, not on the coast, not next to a city and not in a special area where these are waived).
