City Nationality sprites, is it possible to have more than the standard number, for example could I create a Byzantine nationality set with the Hagia Sofia as their church.

Nope. I don't think that's possible. There is no file telling where the city graphics are stored that I have been able to find, thus it's most likely hardcoded.
