Now my race has a -1 sign, I studied the improved signs and now I can’t put it because I can’t remove the green signs

The -1 indicates that the template uses more trait points (cost of positive traits, which you have called green signs) than you're allowed when creating a new template. Having access to such a template is an advantage!

But if you prefer different green signs, you can create a template based on any existing template of the same species, so starting from the weakest (most likely the original) template will give you the most flexibility.

If you take the second bio-ascension, you'll unlock removing positive traits in addition to maximizing available trait points and exclusive positive traits (these traits will have blue signs).

Yeah, -1 means you are ahead of the game. You can "apply" a template even if it wouldn't have been possible for you to create it in the first place.
